The human enclave is the old fortress from an old war, and the titans were that enclave's weapons against the rest of the world. It worked a bit too well, though, they lost control, and now they're trapped, under attack by their own weapons. The human titans are people who used to live in the enclave, but split off during the war when they realized what was happening, and/or didn't want to be used as building materials. The human titans' condition can be passed down through families, but first must be introduced via the serum-thingy, whatever it is.

Not, it's not the best thought-out theory, but it's just a first shot at a theory.

Theres a bit of a hole in that one of the volume jackets had some upside down text explaining the history of the people. If its true and not propoganda the walls were actually FOUND by the ancestors of the current inhabitants who were the rich and powerful of the old world who were able to afford transport along with their servants.

My guess is that it's going to turn out being a lesson on rampant consumption. At least, that seems to be a thematic reveal that it's headed for, what with the constant repetition of the scarcity of resource within the walls (and the strife and death that accompanies that), combined with the mindless, wasteful consumption of the Titans. Suspect #1 is some sort of deity-level creature, #2 is a powerful and wealthy individual or group of humans, #3 is... aliens. Regardless of the cause, though, it will basically be because some entity wanted, either well-meaning or spiteful, wanted to teach humanity about the problems that arise from careless excess.

Apparently the original proposal mini manga that was given out with the dvds or something involved them having been made by an environmentalist group to exterminate humanity, but considering how much most manga change from their proposal prototype manga, it's pretty unlikely to be the same.

Ha,  didn't know about that. But given how strongly the theme is being laid on I expect that it will be at least somewhat related. We know that the Titans are artificial creations by virtue of them breaking the cube-square law and their unnatural body chemistry, so all that remains is determining the source of the Titan serum and the motive for the creation of the non-human Titans. Given that copies of the body of you-know-who's giant Titan are lining the interior of the walls, I think that it's plausible to expect both the human and inhuman Titans to have a common source, given that the walls were simply there rather than built by the people who live within them.
