Nah, tildes at the end of a sentence are used informally to indicate a sing-song manner of speaking.  Pitch going up and down, that sort of thing.

That said, tread lightly, for you tread upon an old wound of this forum

I use it to signify a... playful tone of voice, usually. I'll use them for flirting, for jokes, for being facetious, etc~

Yeah I've used a few times to give it a sing song quality. Never knew it had any connection with japanese punctuation

Also, since we're so close to it, what the hell is hypertilde (It's this:`) used for?

I was under the impression that this thing (') was meant to be used for the 'single quotation marks' where as this thing (`) was originally meant for conjunctions and other somethin`s.

It's used in Japanese subtitles to denote when you hold a note when singing something, so something like "Ohhhhhhhh", becomes, "Oh~". Then it got picked up by other people who wanted to denote being "whimsical".
