I'd like to see a game where everybody has to request replacement half-way through it and the game's players completely change.

EDIT:
Or how about everybody switches with another player (obviously the mafia would only switch with other mafia players) and everybody needs to figure out what's happening while still playing mafia.

Role Rotation Mafia: every day, each person's roles shift to the next person in a chain. You get to pick your own roles, knowing full well that they'll end up in the hands of others. The chain order is always the same but is initially known only to the mod (and, of course, any players with Employment Agent type roles), but canny players will try and figure out the order to work out who has what and who will get what in future rounds.

Tag Team Mafia.

Actually, this reminds me of another idea I had involving two concurrent games that were linked. In that actions in one game affected the other. Perhaps players could even switch places between the games.

...Fringe Mafia.

We had Quantum Mafia, which was very silly and I don't think it finished.

We've had Time Travel Mafia, which I don't even remember doing...

And we've had Parallel Worlds Mafia which looked insane.

Quantum Mafia was an interesting premise. Basing itself off of a few principles of quantum mechanics, players had no set role until "observed." Everything was percentage-based, but the game seemed to peter out, I believe. It was confusing to keep track of.

*She licks her finger and turns a few pages at once.*

Time Travel Mafia was an odd venture to see if you were able to play a game where future days could affect past days. In fact, the town got incredibly lucky, managing to track down the mafia and bring a townie back to life! Toonyman, you were in that one, showing again your lack of defensive ability as the town attacks you to win the game, mostly due to your poor excuse of being a doctor :P! Overall, the experiment could be deemed a failure, as they determined lack of roleflips was pretty detrimental to town. In fact, I believe no game has included no roleflips since then, but I could be mistaken.

*She slams the book close.*

Parallel Worlds Mafia was interesting. Two games of mafia were played concurrently, with power roles able to affect either game. I enjoyed this one, which highlighted Webadict's manipulative nature and emphasizes his relationship with Dakarian. Webadict and Pandarsenic conspire to kill off the other world's mafia, allowing them to have free reign over their own mafia world. A lucky shot on the first night kills one of the mafia of the other world, and the remaining otherworld mafia is taken down eventually. Pandarsenic, having a power role in the other world, manages to help lock down his mafia world, and the game ends in a Pandarsenic-Webadict victory!
