For what its worth, there is a bug with technology, that sometimes (depending on what items you have already I think), it fires without any cooldown period, dealing ~5 times as much damage, changing it from meh to game breakingly good (aka killing satan in 10 seconds without taking any damage).

I wanted to address something lolcats said above.

I almost solely play Isaac. Why? Because all the other characters have a crippling flaw one way or another. Granted, they can be removed, but with the hardness of the game with the DLC (and I suppose the "Everything's Harder now!" achievement), I find even Cain is a liability. Isaac and Mag are the safest characters to play by far, because they're balanced. Cain, Judas and Samson, they're race cars, they leave on the edge for the benefits.

So, I don't think the game requires any wins really, for you to have a chance. Familiarity with the routine (after having played enough to establish one) gets you through levels. That and strangling your curiosity.

You've really got to abuse arcades with Cain.  If you go in with 10 coins, you'll probably come away with 10 keys and 10 bombs more than you would have, and maybe even a blood pack too.  When I play Cain and things don't absolutely implode, I end up with ridiculous numbers of keys and bombs from the cup game.

Also, the difference with pills using Cain is huge as well, by the end of the game you will probably end up with +2 in everything, compared to getting slightly better then even with Issac.

Eh, you do get better pills, but you get crap ones that are strictly bad.  Specifically talking about bombs are key and I found pills.

Apparently, being able to fly through closed arena gates was a bug, and they fixed it. :(

On another note, I achieved "Mamas Boy" and "The Spider Butt" in the same playthrough, which was quite unexpected.
All I have left are most of the character completions, challenge 3, and Platinum God, achievement-wise. (I also have to redo challenge 1, because I didn't get the achievement for it.)
My total playtime has rocketed up to 147 hours.

So ever since the latest patch Sampson is just pointless. Bloody Rage was nerfed and it seems the item drops have changed and there are less of the overpowered items dropping. 
I've been trying for the past two days to try and kill Satan with Sampson and the first few levels are just too hard for his slow, weak arse.

Milk will also do the reverse affect for Sampons for some reason. Making that item competely worthless. I've also not seen a God Room for the whole time I've played since the patch. I'm leaning towards the latest patch either broke them or change their spawning rates to near nothing

On a different note:

Best idea for a video game villain ever.

Well, I picked this up and somehow beat Sheol for the first time. Can't wait to dig into the expansion bosses (I'm avoiding looking at the spoilers on the wiki for this)

Try using the "flashachievements.exe" file in your Binding of Isaac steam folder. I didn't unlock the candle achievement and after using that it did.

Yea, running into a horseman on basement 2 is just a big f u if you haven't gotten good drops.
Try running into a horseman on basement 1.

While playing with a handicap of,"No picking up items from treasure rooms, stores, arcades, libraries, or those thorn-door rooms."

As Cain.

EDIT: I still grab stuff from challenge rooms(also boss challenge rooms), Devil/Angel rooms, regular drops, and boss drops.

Or getting the headless horseman on Cellar 2. Oh god, why.
