I managed to get the twins to half health on my first try, with jungle equipment, no buffs, and no specialized arena (iirc). It gets harder once the firebreathing starts but if you have the mobility they shouldn't be too much of a problem.

An Excalibur (requires Might, which is dropped by the Destroyer himself) can whittle his health down really fast without consuming any ammo, if you're frugal. The flamethrower is also effective, as you said. Using the chainsaw is good too, but only if you have no other replacements for it, since it's hideously low range will get you probed. I say if you've got the stars, use the Starcannon. The stars will drill through without any problems, and deal respectable damage. Cobalt swords are good for minimalists, but you'll likely get killed if you don't keep on your toes.

Breaker blades are good too.

Yeah, Destroyer is by far the easiest of the three to kill, and Skelly Prime is arguably easier than the Twins.

 There is a cheesing trick that involves hellstone, but even with that it's pretty hard to chip away at them before they maul you.

How do poison dart shooters work against bosses?  I may install a set underground and have them firing, to bash on the worm at all times.  Similarly, darts done above jump range could be used to flak-fire at the other bosses.

Curiously, I had a devourer (that's the pre-wall corrupt worm, right?) that encroached on my rotten flesh farm.  It took a poison dart to the body - twice actually, and had part of the body green while the other was normal.  It was rather interesting, and brutally effective at killing it.

Oh yes, worms affected by DoT's are extremely vulnerable to them. I once used a flamethrower on a World Feeder (post-wall corrupt worm), and watched it burn in all its segmented goodness. The damage caused was astonishing.

 Oh cursed flames, how wonderfully you work. And yes, Skeletron Prime can and will take DoT damage from green flames. Cursed bullets should be causing that all the time.

Also if you insist on using the melee helmet, they count as a melee weapon for the record.

Kinda makes sense. You throw them, so some muscle would be required to use them effectively.

This. But it looks cool, so all is well. Oh, and I'm throwing together some (extremely low quality) videos to show the relative difficulties of the three bosses. They'll be complete rubbish, though.  :P
