Hmm... tough ones, I think framerate is definitely a general request instead of a feature so I don't really think it should be there and I think features should be fleshed out before we worry about framerate, that being said I run steadily around 40fps with my entrances waterfall pouring down my stairs and my legions of dwavres laboring but it might affect someone else more harshly. I'd have to say "more underground diversity" since I love the stone system already but I think there needs to be more randomness in finding stuff underground, turn less into dig wherever you can and more into a tread carefully sort of thing, especially around evil areas.

I don't build so much but I can see how mass build must be crucial for some, priorities needs a lot of improvement as well just general de-stupifiying and less walking all the way across my fortress for one stone.

Raw files. Especially more building types for reactions, and improved flexibility in those so that we can make plants/critters/etc easily.

I will have to vote for the job priorities. By being able to define the order of perfered jobs per dwarf or even just in the init, we will have a very powerful but  natural ability to run our fortresses.

2 major issues commonly ruins things when the rough gets going...
(had to whine a bit since I have 2 prime candidates..  ;)))

1 - Job priorities.
2 - Stop dwarven entrance dance when ordered inside.

But since I'm only allowed one, and #2 could possibly be solved with the implementation of #1...

...my vote goes to: #1 - Job priorities.
(With an attached menu where job-priorities could be altered, this of course executed by the "organizer".)

I'd like to vote for all ten options.  I'd love to vote for about four or five.  But I'm going to vote for the issue that's cost more lost fortresses and time, caused more frustration, rendered more game coolness unavailable, and ruined more fun than any other for me.
