That it is possible that the most important thing might vary by country?

True some things would vary, such as what side of the road is being driven on (right side in the US vs left side in the UK), but the most basic stuff to remember wouldn't vary.

Really, legit answer? How to drive. Every aspect of driving. You don't get off with a 9/10 because you are talking about navigating so many pounds of steel at high speeds alongside others.

I think the worst thing to worry about is... Just that... Worrying.
If your not nervous, you'll have a much easier time, so don't let the whole thing get to ya.

Another thing that I personally experienced is manual vs automatic shifting. I had practice with a manual stick shift, but when the day came to taking the test, I couldn't use our car, as it was(and still is) a piece of junk. :P
I advise you to get practice with the type of shifting mechanism of the car you are going to test with first if you are going to use the other type to what you are used to, or better yet, get practice with the exact car you are going to use. It'll make things alot easier.

In my state you are required to take a multiple choice test also. Most of it was easy, obvious stuff, but there were a lot of questions about drinking and driving (blood alcohol limit, how big the fine is for a DUI and other such stuff). I almost failed the test because I never have and never will drink alcohol in my life and so didn't pay attention to that part in drivers ed.

It was easy where I am and I'm pretty sure you will do fine. I did fail the motorcycle test because I had a migraine when I got there, but I just went back the next day and passed it. No big deal.

Wheel steers, seatbelt stays on, pedals for brake, accelerator and clutch, gear stick for changing gears (unless you are lazy and dumb, then it's automatic.) and handbrake for pulling bitching handbrake turns staying in one place.

Check your blind spots before merging, changing lanes, or pulling in past parked cars. This means look over your shoulder, not in the mirror, even if you have those fancy convex mirror extensions. Indicate. For godsakes keep an eye out for stopsigns and (if they have 'em in the US of A) school zones. Check your mirrors before you brake, your blind spots/oncoming traffic before you turn, and make sure you're always in the right gear before you take a corner.

Hope that helps. Either way, I feel your pain, I'm at the same stage of my license. Failed my first test last week; was going flawlessly, until I hesitated too long at a railway crossing so an oncoming car started to turn across in front of me. I was rolling forward a little, and literally as I was pressing the brake, my assessor freaked and stomped the passenger's side brake. Insta-fail. :(

You crazy kids, it's all in the manual. Ohio has A Digest of Ohio Motor Vehicle Laws that covers everything you ought to know for a normal license. The learners permit has a few trick questions and some pure memory questions. They will try to get you on technicalities, so it's good to not only know the information, but tell it apart from similar things. The TvTrope's Law of Conservation of Detail applies too, if it's mentioned more than once, it's more important to know. I also guarantee they will ask what the legal limit is for alcohol, what the jail time and fines are, and if you are dumb enough to answer anything other than waiting the buzz out for getting over it faster.

My brother failed the first time because he forgot to fasten his seatbelt, so don't do that.

Adjusting your seat properly actually helps a lot. If you aren't being mildly annoyed by an uncomfortable or impractical seat, you can concentrate more on driving.
