Eh.

Hitting him would probably work, as long as he didn't hit back. Which no doubt he would. Then, he knows he can hit you, and that when he deems you annoying it's especially okay to do so.
No, if you hit back when he hits you it's sending the message that he most certainly can not.
So long as you don't end up losing. That wouldn't go well.

My brother wouldn't get out of my room when I asked multiple times, and I was trying to sleep. I dragged him out eventually, and now when I ask he leaves.

A good strong shoulder punch worked for me when I was being an annoying brat, but then again my brother is 4 years older than me so it's not like I could've put up a fight.

IronTomato, if this is as bad as you make it out to be, there's absolutely no way you can get your brother to stop pestering you. Pestering you is his favourite thing, and he won't stop doing that if he can do anything about it. If you beat him up, he'll make sure your parents will discourage you from attempting to lay hands on him again. If you ignore him, he'll find ways to force you to stop ignoring him, for example by wrecking your stuff.

A thing you can try to do about this: How about, instead of actually being annoyed, you just act annoyed. Of course it won't be easy to stop being actually annoyed, but you can do it, just keep in mind that all his tricks are getting old. Don't stop acting annoyed, otherwise he'll use new tricks. Do that until you're immune to his pesterings. Now it's your turn to do the ol' psychological manipulation, and that is limited only by your creativity. How about you occasionally just "miss" the part where he annoyed you? Not too often, mind you. On the other hand, you could get really annoyed about things he actually didn't even intend to be annoying. You could also act badly on purpose, being all hammy and stuff. Or how about you annoy him back? Persistence is the key there.

There's a requirement for that though, you actually have to understand him. He can read you like an open book, that's why he's so effective at annoying you. You need to start reading him too. Before you say that he's impossible to understand, being mentally unstable doesn't entail being unpredictable. If his actions aren't what you'd expect from a sane person, then you need to stop trying to interpret them as if he were one.

Finally, if that doesn't work, you can still just endure him until you can move out. Then you can live in peace, make some nerd friends, and it will be so much more satisfying to watch him crash and burn.

Boy, I'll tell you (lookit me sounding so mature). Before my brother got cancer and we went our seperate ways for a couple years, we would fight just like that. My brother would proudly proclaim that it was his "Life's goal to make me as miserable as possible". He would yell dirty jokes into my ear while I was playing games, get a long stick and repediately poke me in the small of my back from a distance trying to provoke me.
And if I ever, ever retaliated? "Moooooooom! [Flabort] hit me!!!!" "[Flabort], you should know hitting never solved anything. You know he's only trying to get a rise out of you, ignore it."

Turns out my parents were right, you shouldn't hit or hit back; if you're brother is annoying you on purpose like this, don't show any outward signs of it bothering you; instead, keep a journal of what he does and how and when it happens, and give access to this journal to a responsible adult.

For me? Spending a couple months learning to be independent while my whole family was in another country away from me for his treatments taught me important lessons for dealing with my problems, while his near-death experience mellowed him waaaay out. It might have been the hallucinatory drugs, though. So my suggestion is take a vacation from your brother.
