The twist? Optimus Prime is actually just misunderstood, and is being his usual self and making a great sacrifice for the greater good: saving the public from having to interact in any way at all with the Princess, who has bewitched much of the kingdom through unknown means and is incredibly annoying to everyone else. You're playing the Hippos. Can you breach Canterlot security and reach the Princess before her big birthday performance? Will you outwit the elusive band of mercenaries known only as "The Little Ponies"? Yes or no, Optimus will not abide the shedding of innocent blood; you can't kill anyone who doesn't attack you, and because the Princess actually has a name you can't kill her, period, without being seen by the audience as evil. You are otherwise free to cause whatever harm you feel is necessary.

There's an additional plot twist not mentioned in the email (it involves Megatron and shenanigans).
