I got a decent amount of river tiles but I have to stumble my way to glass before my second leader does ANYTHING.

Gah. I'm not sure what to do now. Can more than one labor be used to work the land, like me doing something like:

2 labor to irrigate plain 1
3 labor to farm it three times for 9 food? I'd assume that I can't though.

No, each tile can only be worked by one point of labor.

Actually, the watch tower is a city building, not a tile improvement. So, yes.

Is there any rule against moving 100 zefies mini-cities on every tile?
Or building new cities in general? Can I farm within my city at all?

You can't build new cities (or even outposts) yet, you need improved administration technology to do that. As for mini-cities on every tile, well, the tile system isn't really used except for the production radius. New cities and outposts are built outside on the world map.
And, unless you have something like Zrlv's Mushroom Farm building, your city tile can't produce any resources.

Two questions. Actually, more like three questions.

1a: Regarding improvements that give "+1 to X or Y", like the pasture or water wheel: is this a permanent choice made when the improvement is built, or can you choose which extra resource you get each time you work that tile?
1b: Can you build multiple of such improvements to get +1 to each possible resource? (I'm guessing no, but might as well ask.)
2: Can you mix construction materials (e.g. build a "2 wood or 2 stone" structure with 1 wood, 1 stone)?

I forgot to mention that the zefies are prone to comedy. It usually goes away if things get serious.

Oh, and forgot one other: can we split construction across multiple turns (spend 1 labor now + 1 labor next turn to build something that takes 2 labor)?

One labor refines one wool into one unit of clothing. One unit of clothing clothes 10,000 people, raising morale by 1.
