I'm assuming you are saying you don't have access to the top floor? Because if you do, you can still channel it. Otherwise, build an upwards ramp on the lower floor and then remove it.

Or stairs.

To channel, you have to access the tile from a cardinal direction (not diagonally).  This can cause some headaches at times, but you can usually make it happen.

And sometimes a dwarf will gleefully stand on the wrong side of a tile to channel out his only exit from the area.  It's up to you to plan ahead and not let that happen, one way or another.

I do have access to both the floors both I cannot channel the top floor.

Are you sure? The only reason it would be impossible to channel is if it were a constructed floor, and that would be pretty obvious what with the big flashing Cs.

What do you mean by 'can't see' exactly?

Ah - that makes more sense.  But only in that sentence - overall I'm still baffled.

Can you designate the tile to be channeled? Or can you designate it, but then no dwarf will respond???

If you can't even designate that spot, use <k> - what does it say about that spot, about what it is, exactly?

I've seen Dorfs channel diagonally before.

Then describe the situation in some more detail. Or add some screenshots. Also - what does inspe[k]ting the tiles tell you?
I have seen tiles not showing their designation when there was a stone there. Be sure they can access the floor (preferably with stairs as ramps are a bit harder to use), designate to channel, wait, report.

You could avoid that particular problem by constructing ramps below the floor you want gone... but that doesn't explain the problem. But with that little bit of information you provide us with, no one can really help you :\
