I usually don't even have a main shaft. My fortress is a jumbled collection of storage rooms and cramped little hollows with workshops in them.

It seems to me that

takes only 2 Zlevels per cycle, and is thus easier to designate without confusing yourself.

which has the wagon going in a circular path(should be slightly faster than the multiple switchbacks) and requires even less digging(though you have to offset your dig by 3 every other layer)

 Ah, thanks guys. Now I just need to make sure my designations never interfere with the magma pipe and I'll be golden. There will be gratuitous use of up/down stairs from within the workshop and stockpile rooms, but this shaft will be the backbone of it all.
 Also, when I make an awesome tower this shall be used to make sure my Depot is on the bottom of another tower. A tower sealed from the outside except for a bridge on the top level that connects it to another tower.

You should totally have at least one spire that points downwards, contains most of your open-air portions, and never makes direct ground contact.  Support it entirely by skybridges between the other towers, even during construction.

(Easier than it sounds, actually.)

How do you build downwards? I can't seem to find a constructed means of descending a Z-level, or do you need to start on the bottom floor?

I can't help but think that for deep forts you could do something more efficient with multiple paths

Personally I like ramps; they just seem cooler than stairs. Once or twice I've drawn up plans for forts with a centralized spiral of access ramps for maximum efficiency and coolness, but I'm lazy so mostly I just end up with something like this and call it good enough.

You can't build stairs, or anything else for that matter, downward. There's the problem of trying to build a staircase using stone blocks over a pit, which seems infeasible, if not impossible.

Toady is thinking about a solution for this in the next version, though, for all the underground features. A solution, like, a rope ladder, maybe.

I just do a boring old 3x3 up/down staircase all the way to the bottom.

The danger of a dwarf falling is highly overrated - I don't think I've ever had it happen and I play alot.

I've never had it happen either, but I don't normally have one "main shaft" from top to bottom.  I do create "silos" for housing, warehousing, and office/workshops with stairs from top to bottom in the core.

Nowadays, my stairwells are like this:

X+X
+X+
X+X

This allows for dwarves to walk across the stairwell without bumping into those moving up/down it.

I finally got around to setting up autohotkey to do my main shaft for me. I'm thinking about making a script for my housing now too.
