==The "Filesystem: Floor / Ye gods, What did I step into now?" challenge==
Don't ever build stockpiles. Let everything clutter up the floor. No Chasming stuff either. The Goal is to "too deep" the fortress - or just to stay alive as long as possible. (This covers any type of stockpile, including trash. For extra bonus, bring a breeding pair of cats and keep them.)
Things to look out for: Food will rot. Small dead animals and other refuse will stink up the caves. Trading will not be possible. Unless you place your workshops etc. very cleverly expect LONG walking times (and let's not forget clutter).

Here's a challenge I've been thinking of for about a week...  Although, my first ever fortress hasn't died yet(Somehow...) and I refuse to make another until that one's dead.

It's kinda like Challenge #2: The DETA Challenge...  But worse.

The "Papa, papa, I'm huuuungryy" Challenge

The only food/alcohol you can get is from trading.

To be honest, I don't think it's possible.

Therefore... *ahem*

... And from the beginning point building.

The only food/alcohol you can get is from trading, and the initial equipment-picking-point-buying-screen-area-thing.

Wizard of Oz mode: All the walls in your fortress must be replaced with green glass windows\supports. All the floor space in your fortress not used for other things must be paved in green glass. Buildings and furniture must be made of glass. Outdoor roads may only be made of gold bars.

I'm actually doing something close to this, and was considering calling it the "Glassmaster" challenge (not a gold road, though that's a good idea).

It's not really so much a challenge as it is really annoying and really, really ugly.

Edit: Now with pic of said UglyFort:

Edit 2: Really, the neon green glass does nothing for the fort except give me a headache. Too bad its a treeless map, or I'd replace a lot of that with nice, soothing blue clear glass...

You know the "urist" challenge is actually harder than the hermit challenge. With urist she is pretty much BEGGING for humans to invade...... which makes me more to want to try this  :D KILL ALL WHO COMES. MY FORT. NO TOUCH

Heres one - absolutely everything must be built right next to rivers, chasms and magma  :D

I like to call it "Living on the Edge."

new challenge: No doors allowed. This means that any flood is potentially catastrophic

Dwarf made floods can be easily contained with a channel, and I'm pretty sure you could block natural floods from effecting your fortress by building a curving corridor to the river.
Also, building everything 10 spaces east of the river will make you pretty much immune to any trouble this challenge could offer water-wise.

Remaining difficulties:
1) Someone accidentaly being in areas flooded for farming - negated by the fact don't really tend to go to the empty expanses that will be farms. Atleast until they actually are farms.
2) Low quality rooms due to lack of enclosing. Only a trouble with nobles (which you can drown anyway), and actually a benefit once the economy kicks in and everyone becomes homeless.

The Dirty Dozen Challenge
Glacial Map, with no contact barring goblins.
Set the Dwarf cap to 12 in the ini file
Take as much of anything as you like.

Goal: A bloody end.

Note: I recently did this one myself. You'd be surprised at just how quickly endless waves of ratmen can overcome a small group of dwarves. Then again, maybe you wouldn't be.

"Endangered Species"

Start on a map with elephants. Give every dwarf on your fortress the Hunting job. Keeping dwarves from going outside by any means is forbidden.

Easy Goal: Equip all your dwarves in full elephant bone armor.

Hard Goal: Equip all your dwarves in full elephant leather armor.

Here's one I've been trying recently: only one civilian dwarf, rest military. Goal is to kill the demons through force of arms (ie no clever tricks :P).

The trick with that is to get them a non-military legendary skill or two. That way they have the insane stats that they would need to take on the demons.

Well, as the challenge was intended that would be impossible, you're supposed to instantly draft any dwarf (except your civilian) that you get. Still, I think dwarves keep gaining stats even after their weapon skills are maxed, so it should be possible. I think the problem is going to be feeding and outfitting 199 dwarves with just one.  :)

Maybe you'd have to limit immigration somehow...

Well, you are just suppposed to darft them as soon as they arrive, nothing here says you can't order them to be stationed in a room that will somehow be flooded with water after they are locked in it  :)

Not quite a challenge in the same line as the others but should prove fun anyways...

Hedonistic Heaven:
Before nobility gets anything, all other dwarves must have better accomidations. Nobility never gets anything before the 'rabble'. No shared quarters allowed ever. Cannot knowingly endanger or kill any noble. Cannot kill off any immigrants.
