Using the LNP for 43.03 with TWBT produces the same results that milo is describing. Worked fine when I was using 2D mode on 43.05. I'm using a GTX 780.
How about Standard mode in 43.05?
Too much stuff in the starter pack prone to crashing to be of much use in a comparison.

The 64-bit release of Dwarf Fortress v0.43.05 has new SDL libraries. Maybe those are helping with this problem. I'm guessing Milo would have already test this version, though, when he did all his tests.

Actually I totally forgot to test the 64 bit versions. I refuse to play DF without DFHack, so I never downloaded them...

I'll test them and post results, probably on friday.

I tested 43.5 today, and guess what? It works!

It seems that the new SDL has fixed the problem.

Keep in mind that STANDARD (and by extension TWBT) has trouble (eg the game locks up) with the following:

* Resizing or moving the window.
* Using the hotkey to switch between windowed and fullscreen modes.

Alt+Tab works, so if you want to use TWBT or STANDARD you will probably want to use full screen as well.

I use it in windowed mode TWBT so i can easily switch to Dwarf Therapist or the wiki.
The game only locked up a few times when i switched excessively often or during game launch (eg. before the main menu is properly loaded).

Are you using an Nvidia graphics card? Everyone who reported lockups was using an Nvidia card.
