Ascend ASAP. Mutations are wonderful, wonderful things and you can never have enough of them. Plus getting back to where you were is extremely easy with them. Just make sure to save your mutagen at first. 0.1 larva per mutagen is pretty amazing when you're starting out again, but eventually you'll want to spend it.

What matters is how much mutagen and patience you have.  Ascending as fast as possible isn't the best plan.

Ten million or so isn't a terrible start, but if you can persist, you get a third mutation slot for 244 million.  Get some change above that so you can power them up.

Whichever way you go, absolutely don't spend it all at once.  The larva bonus from unspent mutagen is huge early on.

I got to 100 expansions in no time. Guess that's what you get for really wanting those larvae, and swarmlings are cheap, even upgraded.

Actually it was probably just actually using the spells and not trying to get the lepidoptera to 100% energy efficiency like I did.

I just used my first bunch of energy on the things that give you extra energy per second, until I got a whole 1 per sec deal. Then I just focused on my army.
