As per synchronicity, all cards can only be destroyed by cards displaying cured jokers, so this card cannot destroy cards. E3 is still in play.

So, thanks to Synchronicity, this should inverse the effect of every card in play. Including Synchronicity. Which now read "effect that affect all cards now affect a single one". Which mean this card doesn't apply to Synchronicity.

You created a paradox. Well played. Now, how the fuck will I resolve this?

Ultimately, you've got the last call on this, but I'd resolve it like this:

Jokester reverses one card -> Synchronicity applies that to all cards -> Now Synchronicity is inverted for future card effects.

Alternatively, if a card's effects are unclear, you can ignore its rules, as per rule 10.

As rule 10 says I'm the mod  I say we resolve it my way...

Actually, I'm going to apply it another way. This paradox loop only until the random card whose effect is reversed by the jokester is Synchronicity. So now, Synchronicity is inversed, reading "effect that affect all cards now affect a single one". Which mean. Which mean that, starting from now Descent into Madness is in effect. For this turn, [1] notquitethere has to destroy a long lasting card.

Also, thanks to the new Synchronicity, only a single card has to be batman-related this round. As this condition is fullfilled, New Guy and EnigmaticHat can play whatever they want.
