The joke is that it can't be 'got' and is confusing, so that the jokester can insult listeners by calling them gay (as if that was a bad thing). It's a terrible and juvenile joke, hence the thread title.

Jack Daniels is a famous american brand of whiskey founded in the 1800s, Daniel Boone was a famous settler with a similar name, who presumably killed some Native Americans in his time. There's a stereotype about Native Americans being alcoholics.
