Mostly how they act in relation to others. They both want to make the others stronger, to help them survive, but they have very different methods of doing so. Sona believes in nurturing her clients, doing everything she can to making them stronger and giving them a nice, safe time with the power of teamwork. Adrian believes they must make their own mistakes, then learn from them. Find their flaws, and then learn to stop them individually, each person becoming nearly their own individual unit.

And the Mary Sue believes in assimilating everyone and killing everyone who resists.

If we do a parody sue, I want to postpone it until I get a healing Persona with Recarm so I can revive the casualties.
