ALL other access needs to be closed off?

Correct.
To expand on that a little bit, since currently there are no wagons, caravans don't need a three-tile-wide corridor free from anything. They'll waltz right through a door without a hitch or complaint. Hence the suggestions for using drawbridges to secure/isolate the Depot.

Do they set off pressure plates? I've occassionally thought of covering the landscape in a network (unfilled squares of them) of pressure plates linked to doors somewhere to give a whole-map radar but have never had the patience to try to build it.

They don't need a three tile wide path, but they'll still check for it.  And if a three tile path exists from your trade depot to a map edge they can reach, they'll spawn on a spot on the map edge on that path.  Otherwise, they'll spawn on any random map edge they can reach.

Goblin ambushes and thieves set off any pressure plates.  Kobolds don't set off any pressure plates.  Your dwarves and animals set off pressure plates set to be triggered by civilians, but not pressure plates not set to trigger for civilians.  I don't think traders set off any pressure plates at all.  They used to set off civilian pressure plates in 40d, but that behavior seems to have changed.
