I'm planning on deconstructing the door after I make a drainage sewer about 10 levels down, maybe a 20x20 room, that way once the river spills out of that room, I may have enough time to plug the door with a lever-action floodgate.

Yes, it can be plugged. You'll have to set up pumps to clear the water out the hole, drain the flooding waters into a huge pit, and build a wall/door in the gap.
And the pumps have to be powered, so someone won't drown everyone else by knocking off for a snack.

Luckily, it's a river, so you can power them off a waterwheel.
