I voted 40d with graphical tiles (Mayday) but that is because 40d and 40d9 work identically for my computer and many of the external programs (Dwarf Manager, Dtil, Dwarf Companion) had only 40d support at the time.

40d9. Heavily modded, but vanilla tiles and colors.

40d with mods. I tend to use different computers fairly often, so storing a fortress on a usb drive is enough.
