I play windowed with the normal graphics set and have utilities or the wiki open in the background.

Windowed. My monitor's not a good size so it cuts off the edges of most fullscreen programs.

Windowed.  If something with a mod isn't working right I can quckly switch to notepad or something to look for mistakes while the game shuts down. And I just generally prefer windowed mode anyway.

I also use the curses 800x600. I like how nobody else knows what the heck I'm looking at, and the 800x600 font looks much better than the other standard font. (And my vision is bad so the bigger font helps there too.)

I started out full screen then went Windowed, my FPS really suffered in full screen, oh and the squashed deformity got on my nerves (using a 1440x900 Monitor)  so every font is rectangle and stuff unless I put on the blackspace option.

Windowed, so I can multitask.  Things slow down at 100+ dwarves and you need to give it time to run in order to get anything done.

Full screen with black space. (800x600 for the screen, 800x300 for the playable view.) The non-squareness of full screen without black space drives me nuts (yes, 10x12 isn't quite square, but it's a heck of a lot better than 10x24) and I prefer whatever I'm doing to fill the screen.

Windowed, so I can use the internets at the same time.

I like fullscreen, but square fonts and fullscreen don't play nice (either everything gets squashed, or you get blackspace and might as well be windowed) so I used windowed.

If you could change the number of tiles such that fullscreen actually gave you a full screen of normal non-squashed tiles and took up the whole screen I'd use that in a heartbeat.

I run it full-screen according to XP, but that's actually in the window for Parallels, so that I can fiddle on the forums while my fortress burns.

Most games I have issues running windowed, but DF I have issues running full-screen. It sits somewhere just below a full-time interest and above a passing indulgence in attention required, so I find it's ideal to multitask with something else of a similar nature, like browsing forums or chatting on AIM (Pidgin, actually). I use dual monitors so I can have two fullscreen applications open at once, but no current version of Windows allows a fullscreen app like DF to be up while you're clicking or something in the second window by default (I've seen some games counter this effect), so that's another reason why I've ruled fullscreen out. After playing in windowed for so long anyway, playing fullscreen overwhelms me with feelings of general nerdiness for playing a game that uses ASCII for graphics when I've got games like Oblivion and Call of Duty 4 sitting on my desktop.
