OpenGL makes life easier when it comes to supporting multiple resolutions and may reduce CPU load.

Dwarf Fortress's physics probably could get a decent speed boost by using the GPU to handle vector calculations in parallel. Unless somebody has written a simple general-purpose GPU library that works well with a wide variety of hardware, however, doing so probably wouldn't be worth Toady's time.
