For example if I'm pumping into a sealed room can I build enough pressure to make it explode? If not does it build any more pressure at all. For example of I open the flood gates at the bottom will I get a stronger burst of water/magma if the room is full and the pumps are on vs having the pumps off?

Second question can I use a water fall to peer water wheels? I've got just a tiny corner river so I was thinking I'd build. The water wheels under ground and have the water fall on them for power. Would this work?

Pressure does not build up; once an area is full of liquid, it is full of liquid and that is all that occurs.

If you opened flood gates at the bottom of a cistern that had pumps pumping into it, you would get water coming out more quickly than if the pumps were off. That, however, is due solely to the fact that the pumps would immediately start pumping through the now-open gate. Pressure would not have anything to do with it, and in fact it would be just like having the fluid in the pumps' intakes in the cistern rather than separate from it.

Water falls don't power water wheels, though you could have water fall, then flow beneath water wheels to power them.

Not quite accurate here.

What would happen is the layers of water which are sitting on top of the bottommost z level of water will all 'teleport' out the new opening at the bottom, and will come rushing out as a wall of 7/7 water until the water level in the cistern is down to the bottomost Z level,  This will happen nearly instantly, and will do so wether the pumps are on or not.  The pumps may prolong the expansion of the wall of water, but eventually the water would settle down to the lowest Z level of the cistern and will flow out at whatever rate the pumps are feeding it in.

Pumps will however pressurize up to the level of the pump. So if the pump is a z-level higher and pumps down into a cistern below, it will pump up over the top.
