How do i operate flood gates?

You need to connect them to a lever. Build mechanisms in a mechanic's workshop. Build lever, hit "q" and when over the lever, go through the menu to link up a floodgate.

Link the lever to the floodgate. This is done from the lever.
Pull the lever.

You're playing Dwarf Fortress.  There isn't much of a distinction.

You also need a valid route from lever to floodgate while connecting. This requires two mechanisms. (+1 for the lever).
And when placing the floodgate be sure not to seal your dwarf in.

Once I figured out drawbridges, I never built floodgates for any purpose ever again.  If you don't need it to be secure, a wall hatch or door is fine and won't trap dwarves while building it; if you do need it secure (e.g. from building destroyers) then a bridge works just as well, only it takes a bit longer to get it constructed.

Drawbridges also have the advantage of being any size you want them to be within 10x10, rather than only 1x1 - if you want to control a very large flow of liquid, for example in a drowning chamber that you want to be able to cycle quickly, it is imo much much better to use a 2x10 drawbridge (you could make it 1x10 but I like to see whether it's open or closed by looking at it) than any number of 1x1 floodgates.

I have a hard time imagining my "redirecting of the river" would work with drawbridges instead of floodgates. On the other hand, this is DF we are talking about...
